目的 探讨用双向电泳 (Twodimensionalelectrophoresis,2 DE)对大籽蒿花粉提取液蛋白质组分进行分析的可能性及其实用价值。方法 按常规方法提取大籽蒿花粉变应原 ,经SDS 聚丙烯酰胺凝胶电泳及双向电泳对大籽蒿花粉提取液变应原进行蛋白质组分分析。结果 单纯聚丙烯酰胺电泳结果可见有 4个较明显的条带 ,分子量分别为 6 2kD ,5 2kD ,4 3kD和 2 5kD。经双向电泳蛋白组分分析 ,发现大籽蒿花粉提取液有 85种蛋白组分。其中 ,第 139点的分子量为 31.4 9kD ,等电点为 5 .39;第14 3点的分子量为 31.86kD ,等电点为 6 .38。这两个点与文献所报道组分A2c的蛋白质分子量为31kD ,等电点分别为 5 .3和 6 .35一致。结论 双向电泳对大籽蒿花粉提取液蛋白质组分进行分析具有实用价值 ,并可进一步用于变应原致敏组分的研究
Objective To explore the possibility and practical value of two-dimensional electrophoresis (Twodimensionalelectrophoresis, 2 DE) for analyzing the protein components of the pollen extract of Artemisia major. Methods The pollen allergens of Artemisia major were extracted by conventional methods. The protein components of the pollen allergens of Artemisia major were analyzed by SDS polyacrylamide gel electrophoresis and two - dimensional electrophoresis. Results The polyacrylamide gel electrophoresis showed that there were 4 obvious bands with molecular weights of 62kD, 52kD, 43kD and 25kD, respectively. After two-dimensional electrophoresis analysis of protein components, found that the Artemisia major Artemisia pollen extract 85 protein components. Among them, the first 139 points molecular weight of 31.4 9kD, isoelectric point of 5.39; 14th point of the molecular weight of 31.86kD, isoelectric point of 6.38. These two points and the literature reported component A2c protein molecular weight of 31kD, isoelectric point were 5.3 and 6.35 respectively. Conclusion Two-dimensional electrophoresis has practical value for the analysis of protein components of the pollen extract of Artemisia major and can be further used for allergen sensitized components
