目的:从基因角度为新型隐球菌性脑膜炎的快速诊断提供一种新方法.方法:应用聚合酶链反应技术检测和鉴定脑脊液中的新型隐球菌特异性基因.结果:对引物的特异性试验证明具有较高的特异;对引物的敏感性试验可扩增出10个菌细胞;用墨汁复染、分离培养、PCR技术,对15例脑膜炎患者的脑脊液同时进行检测,结果分别为:66.6%、86.6%、93.3%.结论:以新型隐球菌特异性基因片段进行体外扩增,对新型隐球菌性脑膜炎的早期诊断具有重要的临床意义.
Objective: To provide a new method for the rapid diagnosis of cryptococcal meningitis from a genetic point of view. Methods: Polymerase chain reaction (PCR) was used to detect and identify Cryptococcus neoformans-specific genes in cerebrospinal fluid. Results: The specificity test showed that the specificity of the primers was high. Ten bacterial cells were amplified by the sensitivity test of primers. Meanwhile, with the ink dye, isolation and culture, PCR technology, 15 cases of meningitis patients with cerebrospinal fluid For testing, the results were: 66.6%, 86.6%, 93.3%. CONCLUSION: In vitro amplification of Cryptococcus neoformans-specific gene fragments has important clinical significance for the early diagnosis of Cryptococcus neoformans meningitis.
