目的:研究微波消融联合5-氟尿嘧啶瘤内注射治疗结肠癌荷瘤鼠的疗效。方法:取c57雄性小鼠,建立结肠癌荷瘤鼠模型,随机分为对照组、化疗组以及联合组;对照组给予生理盐水处理、化疗组给予5-Fu瘤内注射、联合组给予微波消融联合5-Fu瘤内注射。观察三组小鼠的生存率、测量瘤体直径并计算抑瘤率、取瘤体组织检测相关分子的mRNA含量。结果:化疗组和联合组小鼠的抑瘤率及生存率均高于对照组,联合组小鼠的抑瘤率及生存率均高于化疗组,差异均有统计学意义(P<0.05);化疗组和联合组小鼠的肿瘤组织中Survivin、hTERT、Tspan8、LETM1、GPR56、DDL4和Ephrin-B2的mRNA低于对照组,Caspase3、Caspase9以及PCAF的mRNA含量高于对照组,差异均有统计学意义(P<0.05);联合组小鼠肿瘤组织中Survivin、hTERT、Tspan8、LETM1、GPR56、DDL4和Ephrin-B2的mRNA含量低于化疗组,Caspase3、Caspase9以及PCAF的mRNA含量高于化疗组,差异均有统计学意义(P<0.05)。结论:微波消融联合5-氟尿嘧啶瘤内注射能够更有效的抑制肿瘤生长、提高生存率,同时抑制增殖、侵袭分子的表达并增加促凋亡分子的表达。
Objective: To study the effect of microwave ablation combined with 5-fluorouracil intratumoral injection in the treatment of colon cancer bearing mice. Methods: C57 male mice were used to establish colon cancer bearing mice model and randomly divided into control group, chemotherapy group and combination group. The control group was treated with saline, and the chemotherapy group was given 5-Fu intratumoral injection. The combined group was given microwave ablation Combined 5-Fu intratumoral injection. The survival rate of the three groups of mice was observed, the diameter of the tumor was measured and the tumor inhibition rate was calculated, and the mRNA content of related molecules in the tumor tissue was detected. Results: The tumor inhibition rate and survival rate of chemotherapy group and combination group mice were higher than those of control group. The tumor inhibition rate and survival rate in combination group were higher than those in chemotherapy group (P <0.05) The mRNA expressions of Survivin, hTERT, Tspan8, LETM1, GPR56, DDL4 and Ephrin-B2 in the tumor tissue of the chemotherapy group and the combination group were lower than those of the control group, and the mRNA levels of Caspase3, Caspase9 and PCAF were higher than those in the control group (P <0.05). The mRNA levels of Survivin, hTERT, Tspan8, LETM1, GPR56, DDL4 and Ephrin-B2 were lower than those of the chemotherapy group, and the mRNA levels of Caspase3, Caspase9 and PCAF in the combined group were higher than those in the chemotherapy group Group, the difference was statistically significant (P <0.05). CONCLUSION: Microwave ablation combined with 5-fluorouracil intratumoral injection can inhibit tumor growth more effectively, increase survival rate, inhibit proliferation and invasion molecules and increase the expression of pro-apoptotic molecules.
