目的对羊脆木Pittosporum kerrii根皮的化学成分进行研究。方法运用硅胶、凝胶及RP-HPLC等多种色谱技术进行分离纯化,并根据理化性质和波谱数据鉴定化合物的结构。结果从羊脆木根皮70%丙酮提取物中分离鉴定了3个异苯并呋喃内酯类化合物,分别为5-羟基-6-异戊烯基-异苯并呋喃-1(3H)-酮(1)、5-甲基-6-异戊烯基-异苯并呋喃-1(3H)-酮(2)、5-甲氧基-6-(3-O-乙酰基)-丙酰基-异苯并呋喃-1(3H)-酮(3)。其中化合物1为新化合物,命名为羊脆木素A。并对化合物1进行了细胞毒活性测试,其对NB4、SH-SY5Y、PC3、A549和MCF-7细胞的IC50值分别为3.6、5.2、8.8、5.7和6.0μmol/L。结论化合物1~3均为首次从羊脆木中分离得到,化合物1为新化合物,且表现出细胞毒活性。
Objective To study the chemical constituents of the root bark of Pittosporum kerrii. Methods Silica gel, gel and RP-HPLC and other chromatographic techniques were used to separate and purify the compounds. The structures of the compounds were identified based on the physico-chemical properties and spectral data. Results Three isobenzofuran lactones were isolated and identified from 70% acetone extract from the root bark of sheep crispwood as 5-hydroxy-6-isopentenyl-isobenzofuran-1 (3H) -one (1) 5-Methyl-6-isopentenyl-isobenzofuran-1 (3H) -one -isobenzofuran-l (3H) -one (3). Among them, compound 1 is a new compound named as naringin A. Compound 1 was tested for cytotoxic activity with IC50 values of 3.6, 5.2, 8.8, 5.7 and 6.0 μmol / L for NB4, SH-SY5Y, PC3, A549 and MCF-7 cells, respectively. Conclusion Compounds 1-3 were isolated from sheep brittle wood for the first time. Compound 1 was a new compound and showed cytotoxic activity.
