本文采用改良的次黄嘌呤鸟嘌呤磷酸核糖转移酶(HGPRT)试验,观察人皮角朊细胞铅离子体外染毒的遗传毒性。以1μmol/L的6-硫代鸟嘌呤(6-TG)作为选择剂的浓度。最佳基因突变表达时间为5d。当铅的浓度为6μmol/L时,其T/C值最高。该组与试剂对照组比较有显著差异(p<0.05)。同时采用放射自显影及液闪法对两个指标进行观察:(1)每平方厘米面积上总标记细胞数;(2)标记细胞DNA萃取液放射活性cpm。结果表明上述两项指标间存在显著的正相关(r=0.98)。铅离子对人体皮肤角朊细胞可能有微弱的遗传毒性。
In this paper, a modified hypoxanthine guanine phosphoribosyltransferase (HGPRT) test was used to observe the genotoxicity of lead in human epidermal keratinocytes in vitro. The concentration of 1-mol / L 6-thioguanine (6-TG) was used as a selection agent. The best gene mutation expression time was 5d. When the lead concentration is 6μmol / L, the T / C value is the highest. The group and the reagent control group were significantly different (p <0.05). At the same time using autoradiography and liquid scintillation method of two indicators were observed: (1) per square centimeter of the total number of labeled cells; (2) labeled DNA extract cell radioactivity cpm. The results showed that there was a significant positive correlation between the above two indicators (r = 0.98). Lead ions may have weak genotoxicity on human skin keratinocytes.
