WRKY类转录调控因子在高等植物中构成一个基因超家族,它不但广泛参与了植物对非生物胁迫和生物胁迫的反应,还参与了生长发育及多种代谢途径的调控.本研究构建了一个大豆cDNA文库,采用酵母单杂交的方法,用来自AtNPR1启动子区的W-box对构建的大豆cDNA文库进行筛选,获得了GmWRKY57B基因,该基因全长1033bp,编码299个氨基酸,属于WRKY类转录因子的第Ⅲ组;酵母挽救实验及凝胶阻滞实验均表明,GmWRKY57B能够与W-box特异性结合;酵母体内的转录激活实验表明,GmWRKY57B在酵母细胞中具有转录激活功能;GmWRKY57B在大豆根、茎、叶中均有表达;GmWRKY57B基因在烟草中的过表达能够提高转基因烟草植株的抗旱性,表明GmWRKY57B基因在非生物逆境中有一定的功能.
WRKY transcription regulators constitute a superfamily of genes in higher plants, which not only participate in the plant’s response to abiotic stress and biotic stress extensively, but also participate in the regulation of growth and development and various metabolic pathways.In this study, cDNA library was constructed and the soybean cDNA library was screened by W-box from the AtNPR1 promoter region using the yeast single hybridization method.
