目的采用Box-Behnken响应面法(BBD)优化人参茎叶总皂苷脂质体制备工艺。方法在单因素试验的基础上采用Box-Behnken响应面法,研究了脂药比,磷脂胆固醇质量比,PBS缓冲液pH值对人参茎叶总皂苷脂质体制备的影响。结果通过Box-Behnken响应面法确定的最佳制备条件为:脂药比8.08:1,磷脂胆固醇质量比15.07:1,pH7.44,在此条件下的理论包封率为89.17%。结论确定了人参茎叶总皂苷脂质体的最佳制备工艺,该工艺具有成本低,耗能少,时间短,包封率高等优点。
OBJECTIVE To optimize the preparation of total saponin liposomes from ginseng by Box-Behnken response surface methodology (BBD). Methods Based on the single-factor experiment, Box-Behnken response surface methodology was used to study the effects of lipid-to-drug ratio, phospholipid cholesterol mass ratio and pH value of PBS buffer on the preparation of total ginsenoside liposomes. Results The optimum preparation conditions determined by Box-Behnken response surface method were as follows: lipid-drug ratio 8.08: 1, phospholipid cholesterol mass ratio 15.07: 1, pH7.44. The theoretical entrapment efficiency was 89.17% under these conditions. Conclusion The optimal preparation technology of total ginsenoside liposomes in ginseng leaves and stems was determined. The method has the advantages of low cost, less energy consumption, short time and high entrapment efficiency.
