目的测定甲型肝炎(甲肝)减毒活疫苗H2减毒株P2区基因序列,了解其减毒和细胞适应的分子机制。方法通过逆转录-聚合酶链反应,特异扩增甲肝减毒活疫苗H2株成品疫苗及其亲本毒种P2区VP1/2A、2B、2C基因序列,经测序、比对,分析检测毒力基因VP1/2A、2C的变异情况。结果甲肝减毒活疫苗H2成品疫苗及其亲本毒种毒力基因VP1/2A、2C区毒力相关基因簇均为减毒型特征。结论甲肝减毒活疫苗H2减毒株毒力基因VP1/2A、2C均为减毒型特征,且遗传稳定性良好。
Objective To determine the sequence of P2 gene in H2 attenuated Hepatitis A (A) attenuated live attenuated vaccine and to understand its molecular mechanism of attenuation and cellular adaptation. Methods The gene sequences of VP1 / 2A, 2B and 2C in VP2 of live attenuated attenuated live attenuated vaccine strain H2 and its parental strain were amplified by reverse transcription - polymerase chain reaction (PCR). Sequencing, comparison and analysis of the virulence genes VP1 / 2A, 2C variation. Results The live attenuated HAV vaccine H2 vaccine and its virulence genes VP1 / 2A, 2C district virulence-related gene cluster are attenuated features. Conclusions The attenuated virulence genes VP1 / 2A and 2C of live attenuated HAV vaccine are attenuated and have good genetic stability.
