目的 研究性激素对脑损伤后星形胶质细胞( Ast) 反应及胶质瘢痕形成的影响。方法 采用免疫组化、免疫荧光染色及流式细胞仪( F C M) 计数等方法,观察切除性腺及补充性激素后脑穿刺损伤鼠伤灶周围胶原纤维酸性蛋白( G F A P) 免疫组化染色阳性细胞的形态、数量与比例变化。结果 切除性腺鼠脑损伤后,伤灶周围 Ast 反应较单纯损伤鼠明显,表现为胞体肥大,突起增粗、延长, G F A P 免疫组化染色显著增强。补充性激素后, Ast 反应程度减弱,但未恢复到正常。切除性腺及用性激素治疗前后, Ast 数量与比例无显著变化。结论 性激素能显著影响 Ast 形态变化,但对 Ast的分裂、增殖作用不明显。
Objective To study the effects of sex hormones on astrocyte (Ast) reaction and glial scarring after brain injury. Methods Immunohistochemistry, immunofluorescence staining and flow cytometry (F C M) counting were used to observe the expression of collagen fibrillary acidic protein (G F A P) immunohistochemical staining after excision of gonads and supplemented hormones Positive cell morphology, number and proportion changes. Results After excision of brain injury in sexual gonadal rats, the Ast response around the lesion was more obvious than that of pure injury. The histopathology showed that the soma was hypertrophy, the protuberances were thicker and longer, and the GFA P immunohistochemical staining was significantly enhanced. After the addition of sex hormones, ast response decreased, but did not return to normal. There was no significant change in the number and proportion of Ast before and after gonadal resection and sex hormone treatment. Conclusions Sex hormones can significantly affect the morphological changes of asthma, but have no obvious effect on the division and proliferation of Ast.
