目的从细胞及分子水平探讨参芪扶正注射液对乳腺癌患者免疫功能损害的修复作用及机制。方法77例经病理诊断为乳腺癌的患者分为3组,A组22例仅用手术治疗,B组26例术前化疗加手术,C组29例术前化疗加参芪扶正注射液加手术,另设D组22例为良性肿瘤对照。用流式细胞仪检测各组患者肿瘤组织、引流淋巴结中CD83、CD80、CD86蛋白表达水平。结果A、B、C组癌组织、腋窝淋巴结细胞CD83、CD80、CD86蛋白表达水平比D组明显降低(P<0.05);B组除腋窝淋巴结细胞CD83外均低于A组,C组除腋窝淋巴结细胞CD83外均高于B组(P<0.05,P<0.01),其腋窝淋巴结细胞CD80高于A组(P<0.05)。结论肿瘤及化疗对机体免疫功能产生不良影响,参芪扶正注射液可能参与激活树突状细胞功能,上调共刺激分子水平,帮助修复机体的免疫损伤。
Objective To investigate the repair effect and mechanism of Shenqi Fuzheng Injection on immune dysfunction in breast cancer patients at cellular and molecular level. Methods Totally 77 patients with pathologically diagnosed breast cancer were divided into 3 groups. Group A (n = 22) was treated by surgery only. Group B (n = 26) received preoperative chemotherapy plus operation. Group C (n = 29) received preoperative chemotherapy plus Shenqi Fuzheng injection , Another set of D group 22 cases of benign tumor control. Flow cytometry was used to detect the expression of CD83, CD80 and CD86 in tumor tissues and draining lymph nodes. Results The protein expressions of CD83, CD80 and CD86 in group A, B and C were significantly lower than those in group D (P <0.05). In group B, the expression of CD83 in axillary lymph nodes was lower than that in group A, CD83 in lymph node cells were higher than those in group B (P <0.05, P <0.01), and CD80 in axillary lymph node was higher than that in group A (P <0.05). Conclusions Tumor and chemotherapy have adverse effects on immune function. Shenqi Fuzheng Injection may be involved in activation of dendritic cell function, up-regulation of costimulatory molecules, and repair of immune damage.
