目的:探讨麻杏石甘汤含药血清对3型腺病毒感染的人胚肺成纤维细胞(HELF)之转化生长因子-β1(TGF-β1)、血小板衍生生长因子-BB(PDGF-BB)、肿瘤坏死因子-α(TNF-α)mRNA基因表达的影响。方法:以3型腺病毒分别攻击体外培养的HELF,制备麻杏石甘汤含药血清,作用于该细胞,另设正常细胞组、病毒对照组、利巴韦林组,采用原位杂交法检测不同组细胞TGF-β1、PDGF-BB、TNF-α的mRNA基因表达,进行各组间比较。结果:病毒对照组较正常细胞组之TGF-β1、PDGF-BB、TNF-αmRNA的表达均明显升高(P<0.01),含药血清组对腺病毒攻击后的TGF-β1、PDGF-BB、TNF-αmRNA表达均有显著降低作用(P<0.01)。结论:麻杏石甘汤可下调TGF-β1、PDGF-BB、TNF-α的mRNA表达,这可能是其治疗病毒性肺炎的作用机制之一。
Objective: To investigate the effect of Maxingshigan Decoction-containing serum on the expression of transforming growth factor-β1 (TGF-β1), platelet-derived growth factor-BB (PDGF-BB) , Tumor necrosis factor-α (TNF-α) mRNA gene expression. METHODS: The HELF cultured in vitro was challenged with adenovirus type 3, and the serum containing Maxingshigan decoction was prepared. The normal cell group, virus control group and ribavirin group were also established. The in situ hybridization The mRNA expressions of TGF-β1, PDGF-BB and TNF-α in different groups of cells were detected and compared among groups. Results: The expression of TGF-β1, PDGF-BB and TNF-αmRNA in the virus control group was significantly higher than that in the normal group (P <0.01). The levels of TGF-β1, PDGF- , TNF-αmRNA expression were significantly reduced (P <0.01). Conclusion: Maxingshigan Decoction can down-regulate the mRNA expression of TGF-β1, PDGF-BB and TNF-α, which may be one of its mechanisms in the treatment of viral pneumonia.
