目的探讨+GZ作用后,大鼠心肌组织内内皮素-1(ET-1)和内皮型一氧化氮合酶(eNOS)mRNA表达变化。方法24只雄性Wistar大鼠随机分成对照组、+GZ重复暴露后30min、6h和24h四组,每组6只。对照组大鼠G值为+1GZ,实验组大鼠在动物离心机上经历了3次+10GZ/3min(两次间间隔30min)作用。分别于暴露后30min、6h和24h处死大鼠取心,提取总RNA,用半定量反转录聚合酶链反应(RT-PCR)检测方法检测大鼠心肌组织ET-1和eNOSmRNA表达水平。结果+GZ暴露后30min和6h大鼠心肌组织ET-1mRNA明显升高,eNOSmRNA明显降低,但24h时均恢复正常。结论+GZ暴露可使大鼠心肌组织内ET-1和eNOSmRNA表达发生改变,其在心肌损伤中可能起一定的作用,但这种变化是可逆的。
Objective To investigate the expression of endothelin-1 (ET-1) and endothelial nitric oxide synthase (eNOS) mRNA in myocardium of rats after + GZ treatment. Methods Twenty-four male Wistar rats were randomly divided into control group, GZ + GZ group after repeated exposure for 30min, 6h and 24h. The G value of the control group was + 1GZ, and the rats in the experimental group underwent 3 times + 10GZ / 3min in the animal centrifuge (the interval was 30min between two times). The rats were sacrificed at 30 min, 6 h and 24 h after exposure, respectively. The total RNA was extracted and the expression of ET-1 and eNOS mRNA in myocardium was detected by semi-quantitative reverse transcription polymerase chain reaction (RT-PCR). Results After 30 min and 6 h of GZ exposure, ET-1 mRNA in myocardium was significantly increased and eNOS mRNA was significantly decreased, all of which returned to normal at 24 h. Conclusions + GZ exposure can change the expression of ET-1 and eNOS mRNA in rat myocardium, which may play a role in myocardial injury, but this change is reversible.
