目的:利用内毒素休克模型研究大承气汤对内毒素休克家兔白细胞内游离钙浓度的影响。方法:采用Fura-2/AM 负载及荧光法测定对照组、内毒素休克组和大承气汤治疗组家兔白细胞内游离钙离子浓度。结果:与对照组比较,模型组的白细胞内钙离子浓度有明显升高(P< 0.01);与模型组比较,治疗组白细胞内钙离子浓度明显降低(P< 0.01)。结论:大承气汤可使休克家兔白细胞内游离钙浓度明显降低,这是由于大承气汤抗钙超负荷,减轻其对组织的损伤,从而对细胞功能的调控起到保护作用。
OBJECTIVE: To investigate the effect of Dachengqi Decoction on leukocyte intracellular free calcium concentration in endotoxin shock rabbits using endotoxin shock model. METHODS: Fola-2/AM loading and fluorescence measurements were used to determine the intracellular free calcium concentration in rabbits of the control group, endotoxin shock group and Dachengqi Tang group. RESULTS: Compared with the control group, the intracellular calcium concentration in the leukocytes of the model group increased significantly (P < 0.01). Compared with the model group, the intracellular calcium concentration in the leukocytes of the treated group decreased significantly (P < 0.01). Conclusion: Dachengqi Decoction can significantly reduce the concentration of intracellular free calcium in leukocytes in shock rabbits. This is due to Dachengqi Decoction’s anti-calcium overload, which can reduce the damage to tissues and protect the function of cells.
