目的:为了证实PTH基因在胸腺的表达,明确PTH蛋白在胸腺组织的定位以及胸腺来源的PTH是否受到血钙水平改变的调节。方法:使用RT-PCR、real-timePCR和免疫组织化学的方法比较分析同窝2周龄PTH基因敲除纯合子(PTH-/-)和野生型(WT)小鼠以及1-α羟化酶基因敲除纯合子[1α(OH)ase-/-]和野生型(WT)小鼠的甲状旁腺和胸腺的PTH表达差异。结果:PTH-/-小鼠的甲状旁腺和胸腺无PTH表达;同窝WT小鼠甲状旁腺和胸腺均有PTH表达,但胸腺表达水平较甲状旁腺低。PTH在胸腺上皮细胞表达,而不在胸腺淋巴细胞表达。1α(OH)ase-/-小鼠表现为低钙血症。与正常血钙的WT小鼠相比,PTH基因和蛋白的表达水平在1α(OH)ase-/-小鼠胸腺组织中均明显升高。结论:胸腺组织不仅是产生PTH的辅助器官,而且其PTH表达也受到血钙水平的调节。
OBJECTIVE: To confirm the expression of PTH gene in the thymus and determine whether PTH protein is located in the thymus and whether thymus-derived PTH is regulated by changes in serum calcium levels. Methods: RT-PCR, real-time PCR and immunohistochemistry were used to analyze the relationship between PTH gene knockout homozygotes (PTH - / -) and wild type (WT) mice and 1-α hydroxylase Differences in PTH expression between parathyroid and thymus in gene knockout homozygous [1α (OH) ase - / -] and wild type (WT) mice. RESULTS: No PTH was detected in the parathyroid and thymus of PTH - / - mice. PTH was expressed in both the parathyroid and thymus in the littermates, but the expression of thymus was lower than that in the parathyroid. PTH is expressed in thymic epithelial cells, but not in thymic lymphocytes. 1α (OH) ase - / - mice showed hypocalcemia. The expression of PTH gene and protein were significantly increased in thymus tissue of 1α (OH) ase - / - mice compared with WT mice with normal serum calcium. Conclusion: Thymus is not only an auxiliary organ for the production of PTH, but its PTH expression is also regulated by serum calcium level.
