目的:研究腹腔内注射三氧化二砷(As2O3)对小鼠CO2气腹下肝癌H22转移的影响。方法:昆明鼠40只(清洁级),中腹部穿刺置入1mm套管针,自套管针注入1×106肿瘤细胞后,建立CO2气腹,压力8mmHg,时间30min。术后随机分4组,每组10只,分别腹腔内注入:生理盐水,1ml;As2O3(2mg/kg),1ml;As2O3(4mg/kg),1ml;As2O3(4mg/kg)+肝素(10U/ml),共1ml。气腹后第3、7天测量CD44、VEGF的变化;比较各组生存状态、腹围、体重变化及转移瘤直径。结果:与对照组相比,As2O3组CD44、VEGF表达,转移瘤直径均明显降低。高剂量组比低剂量组效果稍好。As2O3肝素组戳口种植率最低。结论:As2O3对CO2气腹腹腔镜肿瘤生长转移有抑制作用。
Objective: To study the effect of intraperitoneal injection of arsenic trioxide (As2O3) on H22 metastasis in mice with CO2 pneumoperitoneum. Methods: Forty Kunming mice (clean grade) were enrolled in this study. A 1 mm trocar was implanted in the middle abdomen. CO2 pneumoperitoneum was established after 1 × 10 6 tumor cells were injected into the trocar. The pressure was 8 mmHg and the time was 30 min. The rats were randomly divided into 4 groups with 10 rats in each group. The animals were injected intraperitoneally with normal saline (1 ml), As2O3 (2 mg / kg), 1 ml of As2O3 (4 mg / kg) / ml), a total of 1ml. The changes of CD44 and VEGF were measured on the 3rd and 7th day after pneumoperitoneum. Survival status, abdominal circumference, body weight and metastatic diameter were compared between groups. Results: Compared with the control group, the expression of CD44 and VEGF and the diameter of metastatic tumor in As2O3 group were significantly decreased. High-dose group than the low-dose group slightly better effect. As2O3 heparin group pooled the lowest planting rate. Conclusion: As2O3 can inhibit the growth and metastasis of CO2 laparoscopic pneumoperitoneum.
