目的 :了解实验性急性胰腺炎大鼠胰腺腺泡细胞超微结构的改变 ,并探讨雨蛙素致急性胰腺炎的机理。方法 :采用皮下注射超大剂量雨蛙素建立大鼠急性水肿性胰腺炎模型 ,用透射电镜观察了雨蛙素刺激 1h ,3h ,6h ,12h及 2 4h后胰腺腺泡细胞的形态学改变。结果 :①腺泡细胞胞质空泡形成。在雨蛙素刺激后 1小时即可见到 ,随着雨蛙素刺激时间的延长 ,空泡数量显著增加 ,6h达高峰 ;体积亦显著增大 ,空泡之间或空泡与酶原颗粒间互相融合成大空泡。②酶原颗粒数量减少。③在 6小时组变还可见腺泡细胞核染色质边集 ,内质网呈螺环状排列。在2 4小时组 ,上述超微结构改变趋于消失。结论 :雨蛙素刺激导致腺泡细胞内空泡形成 ,消化酶原与溶酶体酶的混合可能在急性胰腺炎的形成过程中起关键作用 ,但腺泡细胞的损伤是可复性的。
OBJECTIVE: To investigate the ultrastructural changes of pancreatic acinar cells in rats with experimental acute pancreatitis and to explore the mechanism of rapamycin-induced acute pancreatitis. METHODS: A rat model of acute edematous pancreatitis was established by subcutaneous injection of super-dose of rabekinin. The morphological changes of pancreatic acinar cells were observed by transmission electron microscopy at 1, 3, 6, 12 and 24 hours. Results: ① acinar cells vacuolar formation. The results showed that the number of vacuoles increased significantly at 6h and reached the peak at 6h with the increase of the stimulating time of the ragweed. The volume also increased significantly, and the vacuoles or the vacuoles and the zymogen granules fused with each other into Big bubble. ② reduce the number of zymogen particles. ③ In 6 hours group also visible acini nucleus chromatin edge set, endoplasmic reticulum spiral ring arrangement. In the 24-hour group, the above ultrastructural changes tended to disappear. CONCLUSIONS: Pretreatment with rasagna causes vacuolization of acinar cells. The combination of digestive enzymes and lysosomal enzymes may play a key role in the formation of acute pancreatitis. However, acinar cell damage is reproducible.
