目的 :观测血管紧张素II(AngiotensinII ,AngII)对体外培养的牛眼小梁细胞 (bovinetrabecularmeshworkcells ,TMcell ) 3H胸腺核苷酸 (3H -thymidine,3H -TdR)掺入率的影响 ,探讨原发性开角青光眼 (primaryopen -angleglaucoma ,POAG)的发病机制。方法 :AngII(1× 10 - 7mol·L - 1及 1× 10 - 8mol·L - 1)以及其I型受体 (an giotensinreceptortypeI,AT1)拮抗剂Losartan孵育TM细胞 ,采用检测 3H -TdR掺入率的方法了解细胞增殖状态。结果 :AngII明显的增加了TM细胞的 3H -TdR摄入率 ,Losartan可部分阻断其增殖效应。结论 :AngII可以诱导体外培养的牛眼TM细胞的细胞增殖率升高 ,AT1受体拮抗剂可以部分阻断此促增殖效应
Objective: To observe the effect of Angiotensin II (AngII) on 3H-thymidine (3H-TdR) incorporation in bovine trabecular meshwork cells (TMEC) Pathogenesis of primaryopen-angleglaucoma (POAG). METHODS: TM cells were incubated with AngII (1 × 10-7 mol·L - 1 and 1 × 10-8 mol·L - 1) and Losartan antagonist of its type I receptor (angiotensin receptor - 1 receptor antagonist), and 3H - TdR incorporation Rate method to understand the state of cell proliferation. RESULTS: AngII significantly increased 3H-TdR uptake in TM cells and Losartan partially blocked its proliferative effect. CONCLUSION: AngII can induce the increase of the cell proliferation rate of bovine TM cells cultured in vitro. AT1 receptor antagonist can partially block the pro-proliferative effect
