为了研究成骨生长肽在体外对成骨细胞样细胞的促成骨作用.将新生大鼠颅盖骨成骨细胞样细胞在体外培养 ,实验组细胞在传第二代后加入成骨生长肽 (1 0 - 8mol/ L) ,对照组不加.细胞传代两次后 ,于第四周起 ,分 3、7、1 0、1 4和 2 1 d收集两组培养的细胞和细胞液 ,检测其中的碱性磷酸酶、酸性磷酸酶、胶原、蛋白多糖及钙含量.结果实验组的细胞和培养液中第 3、7、1 0、1 4、2 1天碱性磷酸酶显著地高于对照组 (P“,”To investigate the regulatory role of osteogenic growth peptide (OGP) in rat osteoblast like cell . The isolated osteoblastic cells from calvaria bone of newborn rats were cultured in vitro. They were divided into two groups,one was added with OGP,the other was added with DMEM as the control.The medium was changed twice a week. On the 3rd、7th、10th、14th 、21th culture day, the activity of alkaline phosphatase、acid phosphatase,the contents of proteoglycan、collagen and Ca in the cells and extracellular matrix were detected.The results showed that , the alkaline phosphatase activities and calcium level in the osteoblastic cells of OGP group on the3rd,7th,10th,14th,21th culture day were significantly higher than those of the control groups (P<0.01~0.05), the collagen level of OGP group were 1.5 times higher than the control (P<0.01). It was concluded that OGP can greatly promote the proliferation of osteoblast like cells and produce more bone tissues.
