本试验用卵巢癌细胞株,采用三磷酸腺苷法(ATP法)分别观察四种化疗增敏剂即三苯氧胺、克罗米酚、环孢菌素A及维拉帕米对顺铂(CDDP)或紫杉醇(Taxol)的体外增效作用。结果显示,各增敏剂在较低浓度(1μg/ml)情况下,单用时均无明显抑瘤作用,但与CDDP或Taxol合用时能增强其在一定剂量下对肿瘤细胞的杀伤作用。各增敏剂在较高浓度(5μg/ml)下,三苯氧胺、克罗米酚及环孢菌素A单用有一定抑瘤作用,此时四药与CDDP或Taxol合用时抑制率增高且高于化疗药物或化疗增敏剂单用时的抑制率。增效作用以三苯氧胺和环孢菌素A最强,故提出可将三苯氧胺或环孢菌素A作为CDDP和Taxol的增效剂用于卵巢癌化疗。另外本文检测该细胞株雌激素受体(FR)为阴性,提示三苯氧胺亦可试用于ER(一)的患者。
This experiment used ovarian cancer cell lines, using the adenosine triphosphate method (ATP) to observe the four chemotherapy sensitizers that are tamoxifen, clomiphene, cyclosporine A and verapamil to cisplatin (CDDP) or paclitaxel ( In vitro synergism of Taxol). The synergistic effect is that tamoxifen and cyclosporin A are the strongest, so it is proposed that tamoxifen or cyclosporin A can be used as a synergist of CDDP and Taxol for ovarian cancer chemotherapy. In addition, the detection of the estrogen receptor (FR) in this cell line was negative, suggesting that tamoxifen can also be used in patients with ER (I).
