目的研究黄连解毒汤抑菌活性的物质基础。方法采用倍比稀释法测定黄连解毒汤总生物碱、总黄酮、总环烯醚萜、水提物4个组分群不同细菌的最低抑菌质量浓度(MICs),并首次对生物碱中盐酸黄柏碱进行了体外抑菌活性研究。结果黄连解毒汤在体外有明显的抑菌活性,且不同组分群的抑菌活性明显不同;总生物碱对受试细菌的抑菌活性最强,对革兰阳性菌的MICs为(8.37±5.05)mg/ml,且对革兰阳性菌的抑菌活性显著强于对革兰阴性菌[MICs为(98.96±32.18)mg/ml]的抑菌活性,但盐酸黄柏碱在最大试验浓度800μg/ml,未表现出明显抑菌活性;水提物的抑菌活性次之,对革兰阳性、阴性菌的MICs分别为(43.53±27.53)、(88.54±39.60)mg/ml;总黄酮、总环烯醚萜对受试细菌的抑菌活性较差,尤以总环烯醚萜的抑菌活性最差。结论黄连解毒汤体外具有明显的抑菌活性,生物碱可能是其抑菌活性的物质基础,而盐酸黄柏碱与其抑菌活性可能无直接关系。
Objective To study the antimicrobial activity of Huanglian Jiedu Soup. Methods The MICs of total alkaloids, total flavonoids, total iridoids and water extracts of different groups of Huanglian Jiedu Decoction were determined by fold ratio dilution method. The MICs of alkaloids in alkaloids Alkaloids were studied in vitro antibacterial activity. Results Huanglian Jiedu Decoction had obvious antibacterial activity in vitro, and the antibacterial activities of different groups were significantly different. The total alkaloids showed the strongest antibacterial activity against the tested bacteria, and the MICs of Gram-positive bacteria were (8.37 ± 5.05 ) mg / ml, and the antibacterial activity against Gram-positive bacteria was significantly stronger than that against Gram-negative bacteria [MICs (98.96 ± 32.18) mg / ml] ml, no obvious antibacterial activity was observed. The antibacterial activity of the water extract was second, and the MICs of Gram-positive and -negative bacteria were (43.53 ± 27.53) and (88.54 ± 39.60) mg / ml, Iridoid antibacterial activity of the tested bacteria is poor, especially the iridoid antibacterial activity of the worst. Conclusion Huanglian Jiedu Decoction in vitro has obvious antibacterial activity, alkaloids may be the material basis of its antibacterial activity, and ciprofen hydrochloride and its antibacterial activity may not be directly related.
