用大鼠胸主动脉制备成保存内皮细胞和去除内皮细胞两种血管条标本,以 Tyrode 液灌流。两种血管条在10~(-7)mol/L NE 或50 mmol/L 高钾作用下显著收缩,5~10 min 后依次加入0.01、0.1、0.25、0.5 mg/ml 的熊胆,每次给药间隔10min。观察到两种标本均显示相似幅度的舒张,这种作用随熊胆浓度的增加而增加。0.5 mg/ml 的熊胆可以完全对抗10~(-7)mol/LNE 所致的收缩,而使高钾所致的收缩张力下降50%左右。熊胆对两种血管条的舒张作用无显著差异(P>0.1)。并证明熊胆对血管的舒张作用不依赖于血管内皮细胞。
The rat thoracic aorta was used to prepare two kinds of vascular stripe specimens for preserving endothelial cells and removing endothelial cells, and perfused with Tyrode solution. Both types of vascular strips contracted significantly at 10~(-7) mol/L NE or 50 mmol/L potassium, and 5, 10 min later, 0.01, 0.1, 0.25, and 0.5 mg/ml bile gallbladder. Dosing interval 10min. Both specimens were observed to show a similar amplitude of diastole, and this effect increased with increasing bear bile concentration. 0.5 mg/ml bear gall can completely counteract the contraction caused by 10~(-7) mol/LNE, and the contraction tension caused by high potassium drops by about 50%. There was no significant difference in the relaxation effect of bear gall on the two vascular strips (P>0.1). It also proved that the relaxation effect of bear gall on blood vessels does not depend on vascular endothelial cells.
