目的研究羟基红花黄色素A(HSYA)对小鼠3T3-L1前脂肪细胞增殖和分化的影响。方法采用MTT法、甘油三酯测定、RT-PCR和双萤光素酶报告基因检测等方法观察HYSA对3T3-L1细胞增殖和分化的影响及其可能的机制。结果 0.01 mg/L~10 mg/L HSYA在4 h~96 h抑制3T3-L1细胞的增殖,120 h促进细胞增殖,并具有浓度依赖性。HSYA减少3T3-L1细胞分化过程中脂质和甘油三酯的含量,增加HSL mRNA的表达和启动子活性。结论HSYA对3T3-L1细胞增殖的影响具有双向性,并通过促进HSL启动子的活性,增加mRNA表达,抑制其分化。
Objective To investigate the effects of hydroxysafflor yellow A (HSYA) on the proliferation and differentiation of 3T3-L1 preadipocytes in mice. Methods The effects of HYSA on the proliferation and differentiation of 3T3-L1 cells and its possible mechanism were observed by MTT assay, triglyceride assay, RT-PCR and dual luciferase reporter assay. Results HSYA at 0.01 mg / L ~ 10 mg / L inhibited the proliferation of 3T3-L1 cells at 4 h ~ 96 h and promoted cell proliferation at 120 h in a concentration-dependent manner. HSYA reduces the lipid and triglyceride content during 3T3-L1 cell differentiation and increases HSL mRNA expression and promoter activity. Conclusion HSYA has a bidirectional effect on the proliferation of 3T3-L1 cells, and enhances the expression of HSL promoter and inhibits its differentiation by promoting the activity of HSL promoter.
