目的:建立反相高效液相色谱法测定制首乌和人参首乌胶囊中大黄素的含量。方法:采用SPHERI-SORB ODS2(150mm×4.6mm,5μm)色谱柱,流动相为甲醇-0.1%磷酸(85∶15),检测波长为254nm,柱温28℃,流速1.0mL.min-1。结果:大黄素进样量在0.03~0.20μg范围内与峰面积线性关系良好(r=0.9996),平均回收率制首乌药材为97.5%,RSD为2.6%;人参首乌胶囊为98.0%,RSD=2.3%。结论:方法简便快捷,准确灵敏,重复性好,可用于原药材制首乌及其制剂人参首乌胶囊的质量控制。
Objective: To establish a RP-HPLC method for determination of emodin in Radix Polygoni multiflori and ginseng Shouwu capsules. METHODS: A HPLC-SORB ODS2 column (150 mm×4.6 mm, 5 μm) was used. The mobile phase consisted of methanol-0.1% phosphoric acid (85:15). The detection wavelength was 254 nm, the column temperature was 28°C, and the flow rate was 1.0 mL.min-1. Results: The linear relationship between the emodin dosage and the peak area was in the range of 0.03~0.20μg (r=0.9996). The average recovery rate of Shouwu was 97.5%, RSD was 2.6%, and ginseng Shouwu capsule was 98.0%. RSD = 2.3%. Conclusion: The method is simple, rapid, accurate, sensitive and reproducible. It can be used for the quality control of Shouwu, the original medicinal material, and its preparation, ginseng Shouwu capsule.
