目的探讨环氧化酶-2(COX-2)、基质金属蛋白酶-9(MMP-9)与血管内皮细胞生长因子(VEGF)在胶质瘤发病中的作用。方法选取正常脑组织8例,脑胶质瘤手术标本55例,采用免疫组化的方法分别检测COX-2、MMP-9与VEGF在脑胶质瘤和正常脑组织中的表达水平,并结合细胞学观察进行对比研究。结果 COX-2、MMP-9和VEGF在正常脑组织中均不表达,而在各病理级别的人脑胶质瘤组织中均有表达,各组间差异有统计学意义(P<0.05);COX-2、MMP-9和VEGF的表达强度随脑胶质瘤病理级别呈递增趋势,且COX-2与MMP-9和VEGF的表达均呈正相关(rs=0.631,rs=0.687;P<0.05)。结论 COX-2、MMP-9和VEGF的表达强度随胶质瘤恶性程度的增加而增加,提示三者可能参与了脑胶质瘤的发生发展过程。COX-2、MMP-9和VEGF在脑胶质瘤的阳性表达之间有一定的相关性,提示脑胶质瘤的发生发展是多个因子相互作用的结果,COX-2可能诱导MMP-9和VEGF的表达而促进肿瘤血管的生成和侵袭转移。
Objective To investigate the role of cyclooxygenase-2 (COX-2), matrix metalloproteinase-9 (MMP-9) and vascular endothelial growth factor (VEGF) in the pathogenesis of glioma. Methods Eight cases of normal brain tissue and 55 cases of glioma were selected. The expression of COX-2, MMP-9 and VEGF in gliomas and normal brain tissues were detected by immunohistochemistry Comparative study of cytology. Results COX - 2, MMP - 9 and VEGF were not expressed in normal brain tissue, but were found in all pathological grades of human glioma tissues. The differences among the groups were statistically significant (P <0.05). The expression of COX-2, MMP-9 and VEGF increased with the pathological grade of glioma, and there was a positive correlation between the expression of COX-2 and MMP-9 and VEGF (rs = 0.631, rs = 0.687; ). Conclusions The expression of COX-2, MMP-9 and VEGF is increased with the malignant degree of glioma, suggesting that the three may be involved in the development of glioma. COX-2, MMP-9 and VEGF in gliomas have a certain correlation between the positive expression, suggesting that the occurrence and development of glioma is a result of the interaction of multiple factors, COX-2 may induce MMP-9 And VEGF expression and promote tumor angiogenesis and invasion and metastasis.
