目的探讨塞来昔布对LoVo移植瘤中环氧化酶-2(COX-2)、血管内皮生长因子(VEGF-C)的表达及对微血管生成的影响。方法移植LoVo的裸鼠分为三组,分别予安慰剂、塞来昔布40 mg/kg和80 mg/kg,第21天取瘤,测定抑瘤率和相对肿瘤增殖率,免疫组化法检测移植瘤中COX-2和VEGF-C的表达,并测定肿瘤微血管密度(MVD)。结果①VEGF-C的表达与COX-2的表达相关,三组中COX-2和VEGF-C的表达差异无统计学意义;②MVD与VEGF-C、COX-2表达相关,给药组MVD低于对照组。结论 COX-2和VEGF-C的表达与肿瘤血管生成有密切关系;塞来昔布可抑制肿瘤新生血管生成。
Objective To investigate the effect of celecoxib on cyclooxygenase-2 (COX-2), vascular endothelial growth factor (VEGF-C) expression and angiogenesis in LoVo xenografts. Methods Nude mice with LoVo transplantation were divided into three groups: placebo, celecoxib, 40 mg / kg and 80 mg / kg respectively. On the 21st day, the nude mice were sacrificed and the tumor inhibition rate and relative tumor growth rate were measured. Immunohistochemistry The expression of COX-2 and VEGF-C in the xenografts was detected and the tumor microvessel density (MVD) was measured. Results ① The expression of VEGF-C was correlated with the expression of COX-2. There was no significant difference in the expression of COX-2 and VEGF-C among the three groups. ② The expression of VEGF was correlated with the expression of VEGF-C and COX- Control group. Conclusion The expression of COX-2 and VEGF-C is closely related to tumor angiogenesis; Celecoxib can inhibit tumor angiogenesis.
