目的探讨亚砷酸钠对人上皮角质形成细胞系(Hacat细胞)PI3K/AKT信号通路中细胞周期相关分子GSK-3β、CyclinD1的影响。方法将人Hacat细胞暴露于浓度分别为0、10、25、50、100μmol/L的亚砷酸钠6 h,采用Western Blot法检测细胞总蛋白的GSK-3β,磷酸化GSK-3β(p-GSK-3β),CyclinD1和磷酸化CyclinD1(p-CyclinD1)的蛋白表达水平。结果与对照组比较,10、25、50、100μmol/L染砷组总蛋白中p-GSK-3β和CyclinD1蛋白表达水平均显著增高(P<0.05),而p-CyclinD1蛋白表达水平显著降低(P<0.05)。结论亚砷酸钠可影响Hacat细胞中的GSK-3β分子,使其磷酸化失活,进而失去对细胞周期相关蛋白CyclinD1的磷酸化作用,从而促进CyclinD1的表达。
Objective To investigate the effects of sodium arsenite on cell cycle related molecules GSK-3β and CyclinD1 in PI3K / AKT signaling pathway of human epithelial keratinocytes (Hacat cells). Methods Human Hacat cells were exposed to sodium arsenite at concentrations of 0, 10, 25, 50 and 100 μmol / L for 6 h. Western Blot was used to detect the expression of GSK-3β, phosphorylated GSK-3β (p- GSK-3β), CyclinD1 and phosphorylation of CyclinD1 (p-CyclinD1). Results Compared with the control group, the protein expressions of p-GSK-3β and CyclinD1 in 10, 25, 50, 100 and 100 μmol / L groups were significantly increased (P <0.05), while p-CyclinD1 protein expression was significantly decreased P <0.05). Conclusion Sodium arsenite can affect the GSK-3β molecule in Hacat cells, inactivate its phosphorylation, and then lose the phosphorylation of CyclinD1 and promote the expression of CyclinD1.
