建立SD大鼠正畸牙移动模型,隔日于正畸牙颊侧牙龈黏膜下单独或联合注射10ng PDGF-BB及200ng IGF-Ⅰ,加力10d后处死大鼠,取材用免疫组织化学方法检测正畸牙压力侧牙周膜细胞中整合素β3的表达。结果:PDGF-BB、IGF-Ⅰ单独或联合应用均能增强压力侧牙周膜细胞中整合素β3表达(P<0.01);与PDGF-BB、IGF-Ⅰ单独应用比较,IGF-Ⅰ加PDGF-BB组牙周膜细胞中整合素β3表达明显增强(P<0.05和P<0.01)。结论:外源性PDGF-BB和IGF-Ⅰ在大鼠正畸牙的局部注射能上调压力侧牙周膜细胞中整合素β3表达,二者联合应用具有协同效应。
The model of orthodontic tooth movement in SD rats was established. On the other day, 10ng PDGF-BB and 200ng IGF-Ⅰ were injected alone or in combination on the buccal side of the gingival mucosa of the orthodontic tooth. The rats were killed 10 days after the force was applied. The specimens were examined by immunohistochemistry Expression of Integrin β3 in Periodontal Ligament Cells in Orthodontic Pressure. Results: Compared with PDGF-BB and IGF-Ⅰ, PDGF-BB and IGF-Ⅰ alone or in combination could increase integrin β3 expression in periodontal ligament cells (P <0.01) -BB group, the expression of integrin β3 in periodontal ligament cells was significantly increased (P <0.05 and P <0.01). Conclusion: Local injection of exogenous PDGF-BB and IGF-Ⅰ in rat orthodontic tooth can up-regulate the expression of integrin β3 in the periodontal ligament cells in the pressure-bearing side. The combined application of them has a synergistic effect.
