DNA分子标记技术是检测遗传差异的一项新技术 ,特别是RAPD标记技术应用更加广泛 ,DNA质量是保证RAPD分析成功的关键。本文采用改良SDS法对油菜叶片总DNA进行了提取 ,经琼脂糖凝胶电泳和紫外光光度计分析 ,结果表明 :1、所提取的DNA浓度和纯度都高 ,可以作为分子生物学研究所用 ,并为本实验后续的PCR扩增奠定了良好的材料基础 ;2、不同时期叶片DNA的含量不一样 ,苗期幼叶DNA含量比青荚期无柄叶DNA含量高
DNA molecular marker technology is a new technology to detect genetic differences. In particular, RAPD marker technology is more widely used and DNA quality is the key to successful RAPD analysis. In this paper, the improved SDS method was used to extract the total DNA of rape leaves. The results of agarose gel electrophoresis and UV photometer showed that: 1, the extracted DNA concentration and purity were high, which could be used as molecular biology research, And laid a good material foundation for the subsequent PCR amplification.2. The content of DNA in leaves at different stages was different, the content of DNA in young leaves at seedling stage was higher than that in leaves without pods
