目的研究2011年吉林省手足口病死亡病例肠道病毒71型的全基因组特征,分析核苷酸及氨基酸变异特点,以探讨变异与致病性可能存在的关联。方法采用分段RT-PCR法对2011年吉林省手足口病死亡病例肠道病毒EV71分离株JL2011-56全基因组进行扩增和测序,与Gen Bank参考株进行核苷酸和氨基酸比对分析,并对全基因组序列进行遗传进化分析。结果 JL2011-56分离株全长7 405 bp,腺嘌呤核苷酸和尿嘧啶核苷酸较为丰富,存在多处氨基酸突变位点。结论吉林省分离株JL2011-56的氨基酸突变位点可能与其致病性相关。
Objective To investigate the genome-wide characteristics of enterovirus 71 death from hand-foot-mouth disease in Jilin province in 2011 and analyze the characteristics of nucleotide and amino acid variation to explore the possible association between mutation and pathogenicity. Methods The whole genome of EV71 strain JL2011-56 from the death of hand, foot and mouth disease in Jilin province in 2011 was amplified and sequenced by segmented RT-PCR. Nucleotide and amino acid alignments were analyzed with Gen Bank reference strains. The whole genome sequence was analyzed by genetic evolution. Results The full length of JL2011-56 strain was 7 405 bp. Adenine nucleotide and uracil nucleotide were abundant, and many amino acid mutation sites existed. Conclusion The amino acid mutation sites of JL2011-56 in Jilin Province may be related to its pathogenicity.
