目的:探讨中药复方固本防哮饮对哮喘缓解期小鼠IκB激酶(IKKβ)及核转录因子(NF-κB)表达的影响。方法:采用卵蛋白(OVA)致敏和呼吸道合胞病毒(RSV)滴鼻诱导激发BALB/c雌性小鼠,建立哮喘缓解期动物模型,随机分为正常组,模型组,固本防哮饮低、中、高剂量组,孟鲁司特钠组及地塞米松组。用Real-time PCR和Western Blot法分别检测肺组织中IKKβ、NF-κBp65 mRNA和蛋白的表达水平。结果:模型组小鼠肺组织中IKKβ、NF-κBp65 mRNA和蛋白表达均显著高于正常组(P<0.01);固本防哮饮低、中、高剂量组,孟鲁司特钠组及地塞米松组小鼠肺组织中IKKβ、NF-κBp65 mRNA和蛋白表达均低于模型组(P<0.05)。结论:哮喘缓解期气道炎症可能与IKKβ、NF-κBp65mRNA和蛋白高表达有关。固本防哮饮在一定程度上能降低IKKβ、NF-κBp65 mRNA和蛋白高表达,提示固本防哮饮能抑制IKK/NF-κB信号通路的活性,从而减轻气道炎症,预防哮喘发作。
Objective: To investigate the effects of Guben anti-asthma drink on IKKβ and NF-κB expression in remission asthmatic mice. Methods: BALB / c female mice were induced by ovalbumin (OVA) sensitization and respiratory syncytial virus (RSV) nasal instillation. Animal models of asthma remission were established and randomly divided into normal group, model group, , Middle and high dose group, montelukast sodium group and dexamethasone group. The expression of IKKβ, NF-κBp65 mRNA and protein in lung tissue were detected by Real-time PCR and Western Blot respectively. Results: The mRNA and protein expressions of IKKβ, NF-κBp65 in the model group were significantly higher than those in the normal group (P <0.01). The Guben anti-asthma drink low, medium and high dose group, montelukast sodium group and The expression of IKKβ, NF-κBp65 in dexamethasone group was lower than that in model group (P <0.05). Conclusion: Airway inflammation may be related to the overexpression of IKKβ, NF-κBp65 mRNA and protein during asthma remission. Guben anti-asthma drink can reduce the expression of IKKβ and NF-κBp65 mRNA and protein to a certain extent, suggesting that Guben anti-asthma drink can inhibit the activity of IKK / NF-κB signaling pathway, thereby reducing airway inflammation and preventing asthma attacks.
