由于肿瘤细胞对高温比正常细胞更敏感,很多实验室和临床工作说明局部加温能抑制肿瘤生长,甚至使肿瘤消退。细胞受高温损伤的可能作用机制,也有多方面的工作进行探索。高温对细胞的损伤作用可因温度高低、作用时间长短和细胞周期、细胞内各种代谢情况不同而不同。为了了解高温对肿瘤细胞作用后的损伤与恢复情况,我们利用人体食管癌上皮细胞株(ECa 109),经42.5℃与44.5℃处理后又回到37℃继续培养,观察细胞的变化。加温后微细结构的变化已有报道,本文报道细胞增殖、细胞分裂、~3H-胸腺嘧啶核苷和~3H-尿嘧啶核苷掺入等的变化。
Because tumor cells are more sensitive to hyperthermia than normal cells, many laboratory and clinical work shows that local warming can inhibit tumor growth and even cause tumor regression. Cell damage by high temperature may be the mechanism of action, there are many aspects of the work to explore. The effect of high temperature on cell damage may be due to the temperature, duration of action and cell cycle, intracellular various metabolic conditions vary. In order to understand the damage and recovery of hyperthermia on tumor cells, we used human esophageal epithelial cell line (ECa 109), after 42.5 ℃ and 44.5 ℃ treatment and then returned to 37 ℃ to continue the culture to observe the changes of cells. Changes in the microstructure after heating have been reported. Changes in cell proliferation, cell division, ~ 3H-thymidine incorporation and ~ 3H-uridine incorporation were reported in this paper.
