目的 探讨角膜中期保存过程中细胞凋亡现象,为改进角膜保存方法提供理论依据。方法 采用MK液、MK+EGF液和Optisol3种保存液保存兔和人角膜,用光镜、电镜及TUNEL标记技术检测细胞凋亡发生情况。结果 3种保存液保存3、5、7天的兔角膜,在上皮、基质及内皮细胞层均可见典型的细胞凋亡发生。保存时间不同,保存液不同,TUNEL阳性细胞数不同。MK液和Optisol液保存4天的人角膜同样可见细胞凋亡发生。结论 细胞凋亡是角膜中期保存过程中细胞丧失的另外一种可能机制;细胞凋亡的发生对角膜中期保存效果存在一定的影响;表皮生长因子在角膜中期保存过程中没有阻止角膜细胞凋亡发生的作用。
Objective To investigate the phenomenon of cell apoptosis in the process of corneal metaphase preservation and provide a theoretical basis for the improvement of corneal preservation method. Methods Rabbits and human cornea were preserved in MK liquid, MK + EGF liquid and Optisol 3 kinds of stock solution. The apoptosis of cells was detected by light microscope, electron microscope and TUNEL. Results The corneas of 3, 7 and 7 days were preserved in 3 kinds of preservation solutions. Typical apoptosis occurred in epithelial, stromal and endothelial cell layers. Different storage time, different preservation solution, TUNEL positive cells in different numbers. Human corneas preserved for 4 days in MK fluid and Optisol fluid also showed apoptotic cell death. Conclusions Apoptosis is another possible mechanism of cell loss in the process of corneal metaphase preservation. The occurrence of apoptosis has some effect on the mid-term preservation of cornea. Epidermal growth factor does not prevent the apoptosis of corneal cells during the mid-corneal preservation Role.
