目的:了解肿瘤细胞对多种化疗药物的耐受即多药耐药(MDR)是否与多药耐药基因(mdrl)的表达增高有关。方法:用人mdrl探针,采用原位杂交的方法分析了30例急性白血病患者mdrl基因表达水平。结果:治疗后未缓解或复发的急性白血病患者的mdrl基因表达水平明显高于治疗前患者(P<0.001)及治疗后完全缓解患者(P<0.01)。结论:mdrl基因的表达增高促进急性白血病临床化疗耐受。mdrl基因表达水平的增高是难治性和复发的急性白血病患者治疗难以缓解的原因之一,无论是初治或治疗后患者mdrl基因表达水平的增高常预示着预后差。
OBJECTIVE: To understand whether tumor cell resistance to various chemotherapeutic agents, ie multidrug resistance (MDR), is associated with increased expression of multidrug resistance gene (mdrl). Methods: The mdrl gene expression levels in 30 patients with acute leukemia were analyzed by in situ hybridization using human mdrl probe. Results: The expression level of mdrl gene in patients with acute leukemia that had not been remitted or relapsed after treatment was significantly higher than that before treatment (P<0.001) and complete remission after treatment (P<0.01). Conclusion: The increased expression of mdrl gene promotes the clinical chemotherapy tolerance of acute leukemia. The increased expression level of mdrl gene is one of the reasons for refractory treatment in patients with refractory and relapsed acute leukemia. The increased expression of mdrl gene in patients with primary or secondary disease often predicts a poor prognosis.
