以砹苯甲酸为中间体,通过活化羧基反应,制得了~(221)At偶联癌胚抗原(CEA)单克隆抗体(McAb)。改进了文献报道的方法,并采用HPLC对偶联物进行了分析和鉴定。方法全程标记率不低于30%,经凝胶色谱Sephadex G_(75)柱分离得到的偶联物注射液比活度可达3.7×10~4Bq/μg McAb。通过测定~(211)At-CEA McAb和~(211)At-在鼠血液和其它组织中的动态曲线,还讨论了偶联物在动物体内代谢的稳定性。
The ~ (221) At conjugated CEA monoclonal antibody (McAb) was obtained by the reaction of activated carboxyl group with benzoic acid as intermediate. The methods reported in the literature were improved and the conjugates were analyzed and identified by HPLC. The labeling rate of the method was not less than 30%. The specific activity of the conjugate injection was about 3.7 × 10 ~ 4Bq / μg McAb separated by gel chromatography on Sephadex G_ (75) column. The stability of the conjugates in animals was also discussed by measuring the dynamic curves of ~ (211) At-CEA McAb and ~ (211) At- in rat blood and other tissues.
