Ⅱ、利福平对卡介菌RNA聚合酶的抑制和草分枝杆菌摄取利福平—~(14)C的生物合成研究已有报导利福平能抑制大肠杆菌的RNA聚合酶,本文进一步采用取自卡介菌的纯化RNA聚合酶研究利福平的作用,并以草分枝杆菌研究抗利福平的机制。实验利用DEAE纤维素柱层析和硫酸铵组分法分离RNA聚合酶,继用甘油梯度密离心法纯化。正式实验是研究利福平对纯化RNA聚合酶活力的抑制作用,结果发现,仅在卡介菌纯化RNA聚合酶与药物保温30分钟后才显示抑制作用,说明利福平作用于RNA聚合酶反应的初期阶段,而不是抑制酶的聚合作
