目的建立DM大鼠模型,待其发展为DN,采用自拟方剂益气养阴活血方对其进行干预,并通过观察实验大鼠各项生化指标、肾组织病理学及肾组织中WT1的表达,研究益气养阴活血方对DN的治疗作用及其有效机制。方法选用清洁级雄性SD大鼠60只,随机分组,除空白组外各组给予链脲佐菌素单次腹腔注射,成膜后不同组给予不同药物处理。6周后观察各组大鼠的一般情况,肥大指数,相关生化指标,PAS染色光镜下观察肾脏病理变化,免疫组化检测肾组织中WT1的表达。结果空白组大鼠的一般情况、肥大指数、各项生化指标及肾组织中WT1的表达情况均有较大差异,各造模组之间也有一定差异。结论益气养阴活血方可以减少尿蛋白,改善脂代谢紊乱,可以抑制DN大鼠的肾小球毛细血管基底膜的增厚、系膜基质增生及足细胞的病变,延缓肾脏病理的进展,可以影响肾组织中WT1的表达。
OBJECTIVE: To establish a rat model of DM, treat it with development of DN, and use the self-made formula Qi-invigorating and Yin-nourishing and blood-activating prescription to intervene. Through observing the biochemical indexes, renal histopathology and expression of WT1 To study the therapeutic effect and its effective mechanism of Yiqi Yangyin Huoxue Recipe on DN. Methods Sixty clean male Sprague-Dawley rats were randomly divided into groups and given a single intraperitoneal injection of streptozotocin in addition to the blank group. Different groups were treated with different drugs after the membrane formation. After 6 weeks, the general situation, the hypertrophy index, the related biochemical indexes of rats in each group were observed. The renal pathological changes were observed by PAS staining and the expression of WT1 in renal tissues was detected by immunohistochemistry. Results The general situation, the hypertrophy index, the biochemical indexes and the expression of WT1 in the kidney tissue of the blank group were all significantly different. There were also some differences among the model groups. Conclusion Yiqi Yangyin Huoxue Fang can reduce urinary protein, improve lipid metabolism disorders, can inhibit DN rat glomerular capillary basement membrane thickening, mesangial matrix proliferation and podocyte lesions, delay the progression of renal pathology, Can affect the expression of WT1 in kidney tissue.
