利用细胞培养技术,研究胰岛素浓度为10、100、1000和10000mU/L时对各组别兔主动脉平滑肌细胞之细胞动力学、形态学变化。结果表明:①胰岛素浓度为10mU/L时,细胞动力学和形态学均无明显变化。②胰岛素浓度≥100mU/L时,细胞明显增殖,且DNA增殖率和胰岛素浓度呈正相关(r=0.53,P<0.01)。③在胰岛素作用下(≥100mU/L),平滑肌细胞呈高电子密度(合成型)及低电子密度(收缩型)二种形态学改变;部份平滑肌细胞内出现脂滴,后者有利于泡沫细胞形成。本文结果提示:降低或避免糖尿病患者的高胰岛素血症,可延缓或减轻动脉粥样硬化的发生与发展。
Cell culture technique was used to study the changes of cell viability and morphology of aortic smooth muscle cells of rabbits at insulin concentrations of 10, 100, 1000 and 10000 mU / L. The results showed that: ① insulin concentration of 10mU / L, the cell dynamics and morphology did not change significantly. ② Insulin concentration ≥100mU / L, cell proliferation, and DNA proliferation rate and insulin concentration was positively correlated (r = 0.53, P <0.01). (3) Under the action of insulin (≥100mU / L), the morphology of smooth muscle cells showed two kinds of morphological changes: high electron density (synthetic) and low electron density (contractile); lipid droplets appeared in some smooth muscle cells, Cell formation. The results suggest that: reduce or avoid hyperinsulinemia in diabetic patients, can delay or reduce the occurrence and development of atherosclerosis.
