目的:建立人结肠癌细胞株裸鼠肝转移模型,研究老鹳草提取物对裸鼠肝转移模型的影响。方法:采用BALB/C裸鼠,Ls174 t人大肠癌细胞株,脾脏切除脾脏种植法建立裸鼠肝转移模型。将36只裸鼠随机分成模型对照组、老鹳草提取物组和羟基喜树碱阳性对照组,给予老鹳草提取物喂养,观察一般情况及肿瘤生长情况;采用巢式逆转录聚合酶链反应(RT-PCR)检测裸鼠循环血中pS2表达;标本作病理组织学检查。结果:①脾脏切除脾脏种植法建立裸鼠结肠癌肝转移模型的成功率为100%,病理组织学证实肝转移癌细胞呈人结肠癌低分化腺癌特征;②老鹳草提取物组裸鼠肿瘤数目及质量明显低于羟基喜树碱组(P<0.05);③老鹳草提取物组裸鼠血中pS2表达为7.598±0.046,明显高于羟基喜树碱组3.794±0.020(P<0.05)。结论:老鹳草提取物可显著降低肝转移裸鼠血中pS2表达,抑制裸鼠结肠癌肝转移的发展,且效果优于羟基喜树碱,可作为大肠癌术后预防复发和肝转移的治疗手段之一。
Objective: To establish a model of human colon cancer cell line metastasis in nude mice to study the effects of geranium extract on liver metastasis in nude mice. Methods: BALB / C nude mice and Ls174 t human colorectal cancer cell line were used to establish the model of hepatic metastasis in nude mice. Thirty-six nude mice were randomly divided into model control group, geranium extract group and hydroxycamptothecin positive control group. Geranium herb extract was fed to observe the general situation and tumor growth. Nested reverse transcriptase polymerase chain reaction The expression of pS2 in circulating blood of nude mice was detected by RT-PCR. The specimens were examined by histopathology. Results: (1) The successful rate of colon cancer hepatic metastasis model was 100% in spleen excision spleen implantation and the histological examination showed that liver metastasis cells showed the characteristics of human colon cancer with poorly differentiated adenocarcinoma. (2) The geranium extract group nude mice (P <0.05); ③The expression of pS2 in blood of nude mice with geranium extract was 7.598 ± 0.046, which was significantly higher than that with hydroxycamptothecin (3.794 ± 0.020, P < 0.05). Conclusion: Geranium extract can significantly reduce the expression of pS2 in the blood of nude mice with liver metastasis and inhibit the development of hepatic metastasis of colon cancer in nude mice, and its effect is better than that of hydroxycamptothecin. It can be used as a prophylactic against recurrence and liver metastasis of colorectal cancer One of the means of treatment.
