目的:探讨肿瘤细胞诱导分化剂六亚甲基二乙酰胺(HMBA)和N-乙酰-1,6-二胺基己烷(NADAH)与人红白血病K562细胞DNA的相互作用和诱导肿瘤细胞分化的机理.方法:通过两步合成,将NADAH偶联到对甲基苯磺酰氯活化的异丙基甘油硅胶上,制备高效液相色谱柱,研究人红白血病K562细胞DNA在NADAH柱上的行为及NADAH及HMBA与人红白血病K562细胞DNA的相互作用.结果:以2mol/L(NH4)2SO4pH7.4的磷酸缓冲液做降盐梯度洗脱时,K562细胞DNA大部分很快被洗脱流出,另有一小部分DNA在(NH4)2SO4浓度降至最低时才流出,NADAH柱对这部分DNA的保留能力极强,即有很强的疏水作用;不同的DNA与NADAH具有不同的疏水亲和力.NADAH柱对DNA的离子效应很弱.结论:HMBA和NADAH在诱导肿瘤细胞的分化过程还对DNA有疏水作用.
OBJECTIVE: To investigate the interaction between tumor cells inducing differentiation of HMBA and NADAH in DNA of human erythroleukemia K562 cells and induce tumor cell differentiation The mechanism. Results: Most of the DNA of K562 cells was eluted quickly when desalting with 2 mol / L (NH4) 2SO4 pH7.4 phosphate buffer, and a small part of DNA was minimized at the concentration of (NH4) 2SO4 When the outflow, NADAH column retention of this part of the DNA strong, that is, has a strong hydrophobic effect; different DNA and NADAH have different hydrophobic affinity. The NADAH column has a weak ionic effect on DNA. Conclusion: HMBA and NADAH also induce hydrophobic interaction with DNA in tumor cell differentiation.
