目的:了解体外培养状态下,NH4+对脑星状胶质细胞谷氨酰胺代谢的影响。方法:将不同浓度NH4Cl加入培养液作用不同时间,分别收集细胞及上清进行测定。结果:NH4+作用后,星状胶质细胞谷氨酰胺合成酶活性增加,细胞内及上清液内谷氨酰胺含量增加,且在一定程度上随着NH4+氨的浓度的增大而增加。然而,当NH4+作用时间达到一定时限(72h)后,酶的活性及细胞内谷氨酰肢的量不再增加,甚至有所下降或减少。结论:星状胶质细胞积极参与NH3(NH3NH4+)的代谢。然而,过高浓度的NH4+作用可影响星状胶质细胞功能,使其对NH4的解毒能力下降。
Objective: To investigate the effect of NH4 + on glutamine metabolism in astrocytes under in vitro culture conditions. Methods: Different concentrations of NH4Cl added to the culture medium for different time, were collected cells and supernatant were measured. Results: After treated with NH4 +, the activity of glutamine synthetase increased in astrocytes, the content of glutamine in cells and supernatant increased, and to a certain extent increased with the increase of NH4 + ammonia concentration. However, when the NH4 + effect time reaches a certain time (72h), the activity of the enzyme and the amount of intracellular glutamine do not increase or even decrease or decrease. Conclusion: Astrocytes actively participate in the metabolism of NH3 (NH3NH4 +). However, excessive concentrations of NH4 + can affect astrocyte function and detoxify NH4.
