本文应用反相高效液相色谱法同时测定3~4月龄、7~8月龄和12~13月龄Wistar大鼠红细胞中三磷酸腺苷(ATP)和辅酶I(NAD)水平.结果表明,ATP分别为:0.810±0.214、0.571±0.157和0.325±0.057μmol/gHb;NAD分别为:0.909±0.137、0.592±0.258和0.622±0.122μmol/gHb.红细胞中ATP含量随增龄显著下降。NAD含量7~8月龄较3~4月龄显著下降,而7~8月龄与12~13月龄大鼠NAD含量比较变化不明显.因此认为,ATP可作为衰老诊断的参考指标。NAD与衰老的关系有进一步探讨的必要。衰老过程中细胞代谢变化的研究可能是探讨衰老机理的—个方面。
In this paper, ATP and coenzyme I (NAD) levels in erythrocytes of Wistar rats aged 3 to 4 months, 7 to 8 months, and 12 to 13 months were measured by reversed-phase high performance liquid chromatography. The results showed that ATP was detected separately. They were: 0.810±0.214, 0.571±0.157, and 0.325±0.057 μmol/g Hb; the NADs were: 0.909±0.137, 0.592±0.258, and 0.622±0.122 μmol/gHb. The ATP content in red blood cells decreased significantly with age. The NAD content of 7 to 8 months of age was significantly lower than that of 3 to 4 months, while the changes of NAD in 7 to 8 months and 12 to 13 months of age were not significant. Therefore, ATP can be used as a reference indicator for the diagnosis of aging. The relationship between NAD and aging is further discussed. The study of changes in cellular metabolism during aging may be an aspect of the study of aging mechanisms.
