目的:建立全蚕粉中槲皮素、山奈素的含量测定方法。方法:采用反相高效液相色谱法(RP-HPLC),色谱柱为KromasiL-C18柱(4.6 mm×250 mm,5μm),柱温为室温,流动相甲醇-0.4%磷酸(50∶50),流速1.0 mL.min-1,检测波长360 nm,进样量10μL。结果:槲皮素和山奈素得到良好分离,分别在0.012 5~0.400 0μg(r=0.999 8),0.014 0~0.448 0μg(r=0.999 9)呈良好的线性关系,平均回收率分别为97.53%(RSD 1.9%),100.3%(RSD 1.0%)。结论:该方法快速准确,可满足定量分析的需要。
Objective: To establish a method for the determination of quercetin and kaempferol in whole silkworm powder. Methods: RP-HPLC was used on a KromasiL-C18 column (4.6 mm × 250 mm, 5 μm). The column temperature was at room temperature and the mobile phase was methanol-0.4% phosphoric acid (50:50) , The flow rate of 1.0 mL.min-1, detection wavelength of 360 nm, injection volume 10μL. Results: Quercetin and kaempferol were separated well, with a good linear relationship between 0.012 5 ~ 0.400 0 μg (r = 0.999 8) and 0.014 0 ~ 0.448 0 μg (r = 0.999 9), with the average recoveries of 97.53% (RSD 1.9%), 100.3% (RSD 1.0%). Conclusion: The method is fast and accurate, which can meet the needs of quantitative analysis.
