甘蔗(Saccharum)作为世界上最重要的糖料和能源作物之一,甘蔗花叶病为甘蔗生产带来很大负面影响。基于此,在海南省儋州两院附近采集具有甘蔗花叶病典型症状的甘蔗叶片,利用超速离心法分离甘蔗花叶病病原粗提纯,置于电镜下观察,发现病原为单一组分,未发现其他病毒病原。为进一步验证其外壳蛋白分子量大小,对病毒粗提纯液进行聚丙烯酰胺凝胶电泳(SDS-PAGE)分析,结果表明:其外壳蛋白分子量约为36 kd,与多数报道的高粱花叶病毒(Sorghum mosaic virus,Sr MV)外壳蛋白分子量接近,初步鉴定儋州两院地区甘蔗花叶病病原为高梁花叶病毒,为建立快速的检测体系及预防措施等提供依据。
Saccharum As one of the most important sugar and energy crops in the world, sugarcane mosaic disease has a great negative impact on sugar cane production. Based on this, we collected sugarcane leaves with typical symptoms of sugarcane mosaic disease in two counties of Danzhou in Hainan Province, and isolated them by ultracentrifugation.
