目的:了解Hedgehog(Hh)-Gli信号通路失调控在肝细胞癌(HCC)发生、发展中的作用及其分子靶向治疗的研究现状。方法:以“Hedge-hog、肝癌/hepatocellular carcinoma(HCC)”为关键词,使用CNKI(1994-2009)、NCBI(1974-2009)生物信息学数据库对已发表的文章进行检索,共检索到英文27篇,中文7篇,排除重复研究和与本研究无关的文献,最终纳入分析27篇文章。结果:研究发现,Hh信号通路不仅调控胚胎时期各胚层组织发育,而且与肿瘤发生、发展密切相关。Gli作为脊椎动物Hh信号通路锌指核转录因子,与Hh通路下游基因的特定序列结合,直接调控Hh信号通路目的基因的转录表达,在Hh信号通路调控中起到枢纽作用。Hh-Gli信号转导通路的异常激活参与HCC的发生及恶性生物学特性的维持。环杷明和(或)Gli-si RNA可以阻断Hh信号通路抑制HCC生长、侵袭和运动潜能。结论:Hh通路的激活是HCC侵袭、发展的重要因素,阻断Hh通路可能是一种新的潜在的HCC靶向治疗策略,具有临床应用价值。
Objective: To investigate the role of Hedgehog (Hh) -Gli signaling pathway in the development and progression of hepatocellular carcinoma (HCC) and its molecular targeted therapy. METHODS: The published articles were searched using the key words “Hedge-hog, hepatocellular carcinoma (HCC)” and CNKI (1994-2009) and NCBI (1974-2009) bioinformatics databases 27 English, 7 Chinese, excluding duplicate studies and literature unrelated to this study, culminating in the analysis of 27 articles. Results: The study found that Hh signaling pathway not only regulate the embryonic development of embryonic tissue, but also with the occurrence and development of tumors are closely related. Gli, a zinc finger nuclear transcription factor of vertebrate Hh signaling pathway, binds to a specific sequence downstream of the Hh pathway and directly regulates the transcriptional expression of the target gene of Hh signaling pathway, playing a pivotal role in the regulation of Hh signaling pathway. Abnormal activation of Hh-Gli signal transduction pathway is involved in the occurrence of HCC and the maintenance of malignant biological characteristics. Cyclopamine and / or Gli-si RNA can block the Hh signaling pathway to inhibit HCC growth, invasion and motor potential. Conclusion: The activation of Hh pathway is an important factor for HCC invasion and development. Blocking Hh pathway may be a new potential HCC targeted therapy strategy with clinical value.
