目的:探讨h1 CaP(h1 calponin,碱性调宁蛋白)及ERK1/2(external signal regulated kinase1/2,细胞外信号调节激酶1/2)在妊娠小鼠子宫平滑肌细胞中总蛋白和磷酸化蛋白的表达变化及与分娩的关系。方法:按妊娠阶段的不同,将小鼠分为未孕组、孕7天组、孕12天组、孕17天组、孕18天组、孕19天组、产时组。应用Western blot检测子宫平滑肌中h1 CaP、磷酸化h1 CaP(ph1 CaP)、ERK1/2、磷酸化ERK1/2(p ERK1/2)的蛋白表达。结果:产时组h1 CaP蛋白表达显著高于未孕、孕7天、孕12天、孕17天组(P<0.01),与孕18天、孕19天组比较差异无统计学意义(P>0.05);ERK1总蛋白表达量产时组与其他各组比较差异无统计学意义(P>0.05);ERK2总蛋白表达量产时组与其他各组比较差异无统计学意义(P>0.05)。ph1 CaP/h1 CaP的比值为产时组显著高于未孕组、孕7天、孕12天、孕17天、孕18天、孕19天组(P<0.01),;pERK1/ERK1的比值产时组与其他各组比较差异无统计学意义(P>0.05);pERK2/ERK2的比值产时组显著高于未孕组、孕7天、孕12天、孕17天、孕18天、孕19天组(P<0.01)。结论:子宫平滑肌细胞中h1 CaP蛋白表达在临产前达高峰,ph1 CaP及p ERK2水平在临产时达最高峰。h1 CaP可通过促进ERK2磷酸化参与分娩发动;h1 CaP对分娩发动的影响不通过ERK1途径。
Objective: To investigate the expressions of h1 calponin and ERK1 / 2 in uterine smooth muscle cells of pregnant mice. Expression changes and the relationship with childbirth. Methods: According to different stages of pregnancy, mice were divided into non-pregnant group, 7-day pregnant group, 12-day pregnant group, 17-day pregnant group, 18-day pregnant group and 19-day pregnant group. The protein expression of h1 CaP, phospho-h1 CaP (ph1 CaP), ERK1 / 2, phospho ERK1 / 2 (p ERK1 / 2) in uterine smooth muscle was detected by Western blot. Results: The expression of hPCaP in labor-producing group was significantly higher than that in non-pregnant, pregnant 7 days, pregnant 12 days and 17-day pregnant group (P <0.01), and there was no significant difference compared with pregnant 18 days and 19-day pregnant group > 0.05). There was no significant difference in the expression of total ERK1 protein among other groups (P> 0.05). There was no significant difference in the expression of total ERK2 protein among other groups (P> 0.05 ). The ratio of ph1 CaP / h1 CaP was significantly higher in labor group than in non-pregnant group (P <0.01). The ratio of pERK1 / ERK1 The ratio of pERK2 / ERK2 in the delivery group was significantly higher than that in the non-pregnant group, with 7 days pregnant, 12 days pregnant, 17 days pregnant, 18 days pregnant, Pregnant 19 days group (P <0.01). Conclusion: The expression of h1 CaP protein in uterine smooth muscle cells reaches its peak before labor, and the levels of ph1 CaP and p ERK2 reach the peak at the time of labor. h1 CaP can be involved in labor by promoting ERK2 phosphorylation; h1 CaP does not affect the onset of labor through the ERK1 pathway.
