目的:研究光动力疗法(Photodynamic Therapy,PDT)联合抗肿瘤药物华蟾素能否产生协同效应,为临床抗肿瘤治疗提供新的治疗手段。方法:用新型光敏剂叶绿素衍生物CPD4联合华蟾素治疗移植性H22肝癌和单一使用PDT及华蟾素作比较,观察抑瘤率及肿瘤细胞形态学变化以判别光敏剂华蟾素协同效应。结果:PDT联合华蟾素组、华蟾素组、PDT组的抑瘤率分别为51.69%、33.49%、26.50%,同时PDT联合华蟾素组可明显改善小鼠整体情况。结论:PDT联合华蟾素能明显增强CPD4对小鼠移植性H22肝癌的光敏杀伤效应,电镜提示杀伤作用主要是导致肿瘤细胞核染色体的崩解,同时能明显改善小鼠整体情况。
Objective: To study whether photodynamic therapy (PDT) combined with anticancer drug Huachansu can produce synergistic effect and provide new treatment means for clinical anti-tumor therapy. Methods: A new photosensitizer, chlorophyll derivative CPD4 combined with cinobufacini, was used to treat transplanted H22 hepatocellular carcinoma, and PDT and Huachansu were compared. The inhibitory rate and the changes of tumor cell morphology were observed to discriminate the synergistic effect of the photosensitizer, Cinobufagin. RESULTS: The tumor inhibition rates of PDT combined with Huachansu, Huachansu, and PDT groups were 51.69%, 33.49%, and 26.50%, respectively. At the same time, PDT plus Huachansu group could significantly improve the overall situation of mice. CONCLUSION: PDT combined with cinobufacini can significantly enhance the photosensitization of CPD4 on transplanted H22 hepatoma in mice. Electron microscopy suggests that the killing effect is mainly responsible for the disintegration of tumor cell nuclear chromosomes, and can significantly improve the overall situation of mice.
