应用斑点杂交和半定量逆转录 多聚酶链反应技术对 3 2例鼻咽癌 (NPC)标本、10例慢性鼻咽炎症粘膜(CINE)标本、NPC细胞株HONE1进行MDM2基因扩增与表达的检测。结果显示 :1例NPC有MDM2基因的扩增 ,NPC细胞株HONE1和 10例NPC有MDM2mRNA高表达 ,CINE无MDM2基因的扩增及mRNA的高表达。MDM2基因的高表达与NPC颈淋巴结转移有关 (P <0 .0 5 ) ,而与NPC的T分期无关。提示MDM2基因在NPC中的主要活化方式是高表达而不是扩增 ,MDM2基因的高表达可能在NPC的发生和转移过程中起着重要作用。
The dot blot and semi-quantitative reverse transcriptase polymerase chain reaction (PCR) were used to detect the amplification and expression of MDM2 gene in 32 NPC specimens, 10 CINE specimens and NPC cell line HONE1. The results showed that: 1 case of NPC has MDM2 gene amplification, NPC cell line HONE1 and 10 cases of NPC have MDM2 mRNA high expression, CINE no MDM2 gene amplification and mRNA high expression. The high expression of MDM2 gene was associated with NPC cervical lymph node metastasis (P <0.05), but not with the NPC T stage. It is suggested that the main activation of MDM2 gene in NPC is high expression rather than amplification, and high expression of MDM2 gene may play an important role in the occurrence and metastasis of NPC.
