目的:优化桑黄子实体总多糖的提取工艺并初步分析其单糖组成。方法:采用苯酚-硫酸法测定总多糖含量。以总多糖提取率为指标,通过单因素试验和正交试验考察提取时间、提取次数、料液比对桑黄子实体总多糖微波提取工艺的影响,利用TLC初步确定不同产地和树种桑黄总多糖的单糖组成。结果:最佳提取条件为加50倍量水提取5次,每次15 min。东北桑树、东北白桦树、甘肃桑树及湖南桑树桑黄中总多糖提取率分别为5.37%,2.21%,3.04%,3.35%,均含有半乳糖和葡萄糖;山东桑树桑黄总多糖提取率2.77%,含有木糖、半乳糖、葡萄糖和乳糖;未知来源桑树桑黄中总多糖提取率3.58%,含有葡萄糖。结论:微波提取法稳定性好、提取率高。东北桑树桑黄中总多糖含量较高,可初步确定桑黄子实体总多糖中均含有葡萄糖。
OBJECTIVE: To optimize the extraction process of total polysaccharide from Phellinus igniarius and to analyze its monosaccharide composition. Methods: The total polysaccharide content was determined by phenol-sulfuric acid method. Total polysaccharide extraction rate as an indicator, single factor test and orthogonal test to investigate the extraction time, the number of extraction, solid-liquid ratio polysaccharides polysaccharide polysaccharide extraction process, preliminary determination by TLC TLC Monosaccharide composition of polysaccharides. Results: The best extraction conditions were 50 times the amount of water extracted five times, each time 15 min. The extraction rates of total polysaccharides in northeast mulberry, northeast birch, mulberry and mulberry Morus alba were 5.37%, 2.21%, 3.04% and 3.35%, respectively, and both contained galactose and glucose. The extraction rate of total polysaccharides from Morus alba was 2.77% , Contains xylose, galactose, glucose and lactose; unknown source Mulberry Pine total polysaccharide extraction rate of 3.58%, containing glucose. Conclusion: Microwave extraction has good stability and high extraction rate. Northeast mulberry yellow total polysaccharide content is higher, can be initially determined fruiting body total polysaccharide contains fructose.
