目的研究中华芦荟多糖(AP)对X射线照射后人非瘤细胞株C.Liver和293细胞周期进程及细胞周期相关蛋白表达的影响。方法流式细胞仪检测细胞周期,Westernblot检测细胞周期相关蛋白的表达。结果辐射后293和C.Liver细胞均出现明显的G2/M期阻滞,G0/G1期细胞比例显著下降,50μg/mlAP能显著改变上述趋势。AP预处理能显著下调辐射所致C.Liver细胞P53蛋白水平,显著升高P21、CyclinB1、pRb蛋白的表达,对P27、CDK4和CyclinD1蛋白表达水平无显著影响。结论AP对X射线辐射后非瘤细胞周期紊乱的改善作用与对周期相关蛋白表达的调节有关。
Objective To investigate the effects of aloe polysaccharides (AP) on cell cycle progression and cell cycle related protein expression in human non-neoplastic cell lines C. Liver and 293 after X-irradiation. Methods The cell cycle was detected by flow cytometry, and the expression of cell cycle-related proteins was detected by Western blot. Results Both 293 and C.Liver cells showed significant G2/M arrest after irradiation, and the proportion of cells in G0/G1 phase decreased significantly, and 50 μg/ml AP significantly changed the above-mentioned trend. AP pretreatment could significantly down-regulate the level of P53 protein in C.Liver cells induced by radiation, significantly increase the expression of P21, CyclinB1, and pRb proteins, but had no significant effect on the expression levels of P27, CDK4, and CyclinD1 proteins. Conclusion The improvement of AP cell cycle after X-ray irradiation is related to the regulation of the expression of cycle-related proteins.
