目的检测转化生长因子(TGF)-β1对乳腺癌细胞株增殖能力和T淋巴细胞免疫的影响,探讨TGF-β1在乳腺癌演进过程中的作用。方法噻唑蓝(MTT)比色法检测TGFβ1对乳腺癌细胞株MCF7、MDAMB157、SKBR3和MDAMB453增殖的影响,探求其调控细胞生长的浓度效应和时间效应。酶联免疫吸附试验(ELISA)检测TGFβ1作用于体外培养的外周血单个核细胞(PBMC)前后白细胞介素(IL)2和干扰素(IFN)γ的分泌。结果作用72h内TGFβ1对乳腺癌细胞有增殖抑制作用(P<0.05),不同的浓度(1.0~100.0)μg/L产生的效果不同。96h后效应减弱至消失。在TGF-β1作用下,PBMC培养上清中IL2(296.5±79.4)ng/L和IFNγ水平(665.3±107.0)ng/L明显低于无TGFβ1作用组(644.9±105.5)ng/L和(922.3±184.1)ng/L,(P<0.01)。结论TGFβ1对乳腺癌细胞的增殖有短暂抑制作用,在一定浓度范围内呈剂量依赖性,作用时间有限。TGF-β1能降低T细胞的细胞因子释放,抑制细胞免疫。
Objective To investigate the effect of transforming growth factor-β1 (TGF-β1) on the proliferation and T lymphocyte immunity in breast cancer cell lines and to explore the role of TGF-β1 in the development of breast cancer. Methods The effect of TGFβ1 on the proliferation of breast cancer cell lines MCF7, MDAMB157, SKBR3 and MDAMB453 was detected by MTT assay, and the concentration effect and time effect of TGFβ1 on the growth of breast cancer cell lines MCF7, MDAMB157, SKBR3 and MDAMB453 were explored. The secretion of interleukin (IL) 2 and interferon (IFN) γ was measured by enzyme-linked immunosorbent assay (ELISA) before and after treatment of TGFβ1 on peripheral blood mononuclear cells (PBMCs). Results The effect of TGFβ1 on the proliferation of breast cancer cells was inhibited within 72h (P <0.05). The effects of TGFβ1 at different concentrations (1.0-100.0 μg / L) were different. After 96h weakened to disappear. The levels of IL2 (296.5 ± 79.4) ng / L and IFNγ (665.3 ± 107.0) ng / L in PBMC culture supernatants were significantly lower than those in the absence of TGFβ1 (644.9 ± 105.5) ng / L and (922.3) ± 184.1) ng / L, (P <0.01). Conclusion TGFβ1 has a transient inhibitory effect on the proliferation of breast cancer cells in a dose-dependent manner with limited time. TGF-β1 can reduce the release of cytokines T cells, inhibit cellular immunity.
